Defined Bioscience’s HiDef® ITS Cell Culture Supplement is a chemically defined, animal-origin-free serum replacement used to maintain and support adherent cells under reduced-serum or serum-free conditions. It combines three critical components normally supplied by serum—insulin, transferrin, and sodium selenite—each optimised to sustain cell metabolism, attachment, and viability across a wide range of primary, immortalised, and stem cell types.
Insulin stimulates the uptake of glucose, amino acids, and ions to promote cell growth and prevent apoptosis. Transferrin serves as a high-affinity iron carrier and antioxidant, reducing toxicity associated with free iron exposure. Sodium selenite contributes antioxidant protection and acts as a cofactor for essential selenoenzymes. Together, these factors maintain physiological balance in nutrient-limited conditions and allow foetal bovine serum (FBS) to be reduced to four per cent or less without compromising performance.
HiDef ITS is supplied as a 100× liquid supplement in Earle’s Balanced Salt Solution (EBSS) without phenol red and prepared using fully animal-free components. It is compatible with a wide range of basal media, including Defined Bioscience’s DMEM/F12, and is validated through fibroblast outgrowth and viability testing. Each 10 mL vial is sufficient to prepare one litre of complete medium.
