ECMatrix-511 E8 Laminin Substrate is a xeno-free, ready-to-use recombinant laminin-511 E8 fragment designed for feeder-free culture of human pluripotent stem cells (ES and iPS cells). It supports stable maintenance and efficient expansion by binding to the α6β1 integrin expressed on pluripotent stem cells, providing a defined and animal-free surface for consistent cell culture.
This formulation (Cat. No. CC160100ML) is produced in CHO-S cells under serum-free conditions, ensuring consistent, animal-free performance. It is supplied as a ready-to-use liquid at 1.6 μg/mL in recombinant human serum albumin (HSA) and phosphate-buffered saline (PBS), eliminating the need for plate precoating. The substrate undergoes rigorous quality control, including purity (>95% by SDS-PAGE), endotoxin (≤750 EU/mg), sterility, mycoplasma testing, and integrin binding affinity confirmation (10 nM).
Key features include a xeno-free and animal-free format with lot-to-lot consistency; no plate precoating required, saving time during passaging; support for single-cell passaging without ROCK inhibitor, enabling CRISPR editing or clonal isolation; higher adhesion and growth rates; and easy handling with no chilling of consumables required.
Store at 2–8°C. Protect from light and avoid multiple freeze-thaw cycles. For coating, dilute to 0.25 μg/cm² and incubate for 1 hour at 37°C, 3 hours at room temperature, or overnight at 4°C. Remove remaining fluid before use; do not rinse. Detach cells into small clumps using Accutase or a similar dissociation reagent. For research use or further manufacturing. Not for diagnostic use or direct administration into humans or animals.
