Defined Bioscience’s HiDef® PBS/EDTA Cell Dissociation Reagent is a defined, protein-free formulation for gentle and consistent detachment of adherent cells. It enables routine aggregate passaging or single-cell dissociation without damaging cell surface proteins or compromising viability, making it suitable for pluripotent stem cells, primary cells, and other sensitive adherent cultures.

The reagent consists of Dulbecco’s Phosphate Buffered Saline (DPBS) without calcium or magnesium, supplemented with 0.5 mM EDTA at physiological pH 7.4. This composition facilitates cell detachment through chelation while preserving functionality, reducing stress, and maintaining morphology. HiDef PBS/EDTA can also be used as a preparatory wash step prior to enzymatic digestion, such as trypsinisation.

Each lot is validated using human induced pluripotent stem cells (iPSCs) cultured in HiDef® B8 medium to confirm reliable detachment and recovery. Supplied ready-to-use at 1× concentration in sterile 125 mL bottles, the reagent can be stored at 2–30 °C for up to 18 months. It offers a cost-effective, enzyme-free alternative for applications requiring high post-passaging viability and minimal impact on cell physiology.
